地塞米松对内毒素休克大鼠脑组织细胞因子含量的影响

Effect of Dexamethasone on Cytokine in Brain of Rats with Endotoxin Shock

摘要: 目的探讨地塞米松对内毒素休克大鼠脑组织细胞因子含量的影响。方法54 只健康成年Sprague-Dawley 大鼠分为假手术组、脂多糖组和地塞米松组,后两组静脉注射脂多糖8 mg/kg,地塞米松组于造模成功后10 min 静注地塞米松5 mg/kg。动态监测血压。分别于造模后1 h、3 h、6 h 用ELISA法检测脑组织中白细胞介素(IL)-4、IL-10 和肿瘤坏死因子(TNF)-α的含量。结果脂多糖组与假手术组比较,随着血压的下降,脑组织中IL-4、IL-10 浓度减少(P<0.01),TNF-α增加(P<0.01);与脂多糖组比较,地塞米松组TNF-α含量减少(P<0.01),IL-4、IL-10 含量升高(P<0.01)。结论地塞米松能调节脂多糖诱导的内毒素休克大鼠脑内炎性因子,发挥脑保护作用。

Abstract: Objective To explore the effect of the dexamethasone on the levels of cytokine in the brain of endotoxin shock rats. Methods 54 Sprague-Dawley rats were randomized to the sham group, LPS group (lipopolysaccharide 8 mg/kg, i.v.) and DEX group (dexamethasone 5 mg/kg, i.v., in addition). The blood pressure was measured dynamically. The contents of interleukin (IL)-4, IL-10 and tumor necrosis factor (TNF)-α were detected with ELISA 1 h, 3 h, 6 h after shock. Results As the occurrence and development of shock, blood pressure went down gradually, IL-4 and IL-10 decreased (P<0.01), and TNF-α increased (P<0.01) in LPS group, while the TNF-α decreased (P<0.01), the IL-4 and IL-10 increased (P<0.01) in the DEX group compared with the LPS group. Conclusion Dexamethasone may protect the brain from the endotoxin shock injury in rats through regulating the inflammatory factor.
