电针预处理对大鼠脑缺血再灌注损伤后炎性因子及细胞凋亡的影响①

摘要

目的探讨电针预处理对大鼠脑缺血再灌注损伤后白细胞介素(IL)-1β、IL-6 含量及细胞凋亡的影响。方法36 只清洁级雄性Sprague-Dawley 大鼠按随机数字表法分为假手术组、模型组和电针预处理组,每组12 只。大脑中动脉阻塞(MCAO)建立脑缺血2 h 再灌注模型。再灌注后24 h,酶联免疫吸附法检测血清、脑组织IL-1β、IL-6 的含量,TUNEL染色检测缺血半暗区神经细胞凋亡。结果与模型组比较,电针预处理组大鼠血清、脑组织IL-1β、IL-6 的含量明显下降(P<0.01),TUNEL阳性细胞数显著下降(P<0.001)。结论电针预处理降低炎症反应,减少细胞凋亡。

Abstract

Objective To investigate the effect of electro-acupuncture pretreatment on content of interleukin (IL) -1β and IL-6 in serum and ischemic penumbra, and apoptosis in ischemic penumbra in rats after cerebral ischemia- reperfusion injury. Methods Thirty- six male Sprague-Dawley rats were randomly divided into sham group (n=12), model group (n=12), and electro-acupuncture pretreatment group (n= 12). The middle cerebral arteries were occluded for 120 minutes and reperfused. Twenty-four hours after reperfusion, the level of IL-1β and IL-6 in serum and brain tissue was detected with enzyme-linked immunosorbent, and apoptosis of ischemic penumbra was detected with TUNEL. Results The content of IL-1β and IL-6 in the serum and brain tissue, and the number of TUNEL-positive cells decreased in the electro- acupuncture pretreatment group compared with that in the model group (P<0.01). Conclusion Electro-acupuncture pretreatment may inhibit inflammatory response and apoptosis in cerebral ischemia-reperfusion injury.
