摘要

目的观察慢性应激抑郁模型大鼠血浆凝血功能变化以及电针的干预作用。方法雄性Sprague-Dawley大鼠32只,适应性饲养1周后,随机分成空白组、模型组、氟西汀组、电针组,每组8只。后三组采用慢性应激刺激结合孤养21 d建立大鼠抑郁模型,氟西汀组、电针组分别同时采用氟西汀、电针进行干预。测定各组大鼠凝血酶原时间(PT)、活化部分凝血活酶时间(APTT)、凝血酶时间(TT)、纤维蛋白原(FIB);酶联免疫吸附测定检测大鼠6酮前列腺素F (6-keto-PGF)、P-选择素、血栓烷素B2 (TXB2)的变化。结果模型组旷场试验水平运动次数和垂直运动次数较空白组明显降低(P<0.01),电针组和氟西汀组较模型组明显增加(P<0.01)。与空白组比较,模型组PT、APTT、TT明显缩短(P<0.01),FIB含量升高(P<0.05);血清6-keto-PGF含量降低(P<0.05),P-选择素、TXB2含量明显升高(P<0.01);与模型组比较,氟西汀组、电针组PT、APTT、TT延长(P<0.05),血清6-keto-PGF含量升高(P<0.01), P-选择素、TXB2含量降低(P<0.05),电针组FIB含量降低(P<0.05)。结论慢性应激抑郁模型大鼠凝血功能增强,血小板活化功能异常,电针和氟西汀可改善这些变化。

Abstract

ObjectiveTo observe the change of coagulation function in chronic stress depression model rats and the effects of electroacupuncture (EA) on it. MethodsA total of 32 Sprague-Dawley rats were randomly divided in control group (n=8), model group (n=8), EA group (n=8) and fluoxetine group (n=8) after a week of acclimation. The 21-day unpredictable mild stress combined with solitarily feeding was used to make the depression model in the latter three groups, meanwhile EA group and the fluoxetine group accepted EA and fluoxetine. They were measured the prothrombin time (PT), activated partial thromboplastin time (APTT), thrombin time (TT) and fibrinogen (FIB). Serum levels of 6-keto-prostaglandin F (6-keto-PGF), P-selectin, thromboxane B2 (TXB2) were determined with enzyme-linked immunoabsorbent assay. ResultsThe cross and rear scores were significantly lower in the model group than in the control group (P<0.01). The scores increased in EA group and the fluoxetine group (P<0.01) compared with the model group. Compared with the control group, PT, APTT and TT decreased (P<0.01), FIB increased (P<0.05), the level of 6-keto-PGF decreased, the level of P-selectin and TXB2 increased (P<0.01) in the model group. Compared with the model group, PT, APTT and TT increased (P<0.05), the level of 6-keto-PGF increased (P<0.01), and the level of P-selectin and TXB2 decreased (P<0.05) in EA group and the fluoxetine group, and FIB decreased in EA group (P<0.05). ConclusionThe chronic stress depression rats are impaired in the coagulation function and platelet activation, and can be allayed with electroacupuncture and fluoxetine.
