运动预处理对脑缺血再灌注大鼠缺血半暗区细胞凋亡及相关蛋白P53 表达的影响①

Effect of Exercise Preconditioning on Apoptosis and Expression of P53 in Ischemic Penumbra in Rats after Cerebral Ischemia-Reperfusion

摘要: 目的探讨运动预处理对脑缺血再灌注大鼠脑组织缺血半暗区细胞凋亡程度及P53 蛋白表达的影响。方法36 只雄性Sprague-Dawley 大鼠随机分为假手术组、模型组、运动预处理组,每组各12 只。采用改良Longa 线栓法制备大鼠大脑中动脉阻塞模型(MCAO)。TUNEL法观察缺血半暗区细胞凋亡情况;Western blotting 检测缺血半暗区P53 蛋白的表达。结果脑缺血再灌注24 h 后,模型组TUNEL阳性细胞数较假手术组明显增加(P<0.01),运动预处理组较模型组明显降低(P<0.01);模型组P53 蛋白表达明显高于假手术组(P<0.01),运动预处理组明显低于模型组(P<0.01)。结论运动预处理可下调脑缺血再灌注大鼠缺血半暗区P53蛋白的表达,有效抑制缺血半暗区皮层细胞凋亡,发挥脑保护作用。

Abstract: Objective To explore the effect of exercise preconditioning on apoptosis and expression of P53 protein in ischemic penumbra in rats after cerebral ischemia-reperfusion. Methods Thirty- six Sprague-Dawley rats were randomly divided into sham group, model group and exercise preconditioning group, with twelve rats in each group. The middle cerebral artery occlusion (MCAO) model was established with modified Longa's method. TUNEL method was used to observe the apoptosis of neural cells in the ischemic penumbra. Western blotting was used to detect the expression of P53 protein in the ischemic penumbra. Results Twenty-four hours after cerebral ischemia-reperfusion, the number of TUNEL positive cells was more in the model group than in the sham group (P<0.01), and was less in the exercise preconditioning group than in the model group (P<0.01). The expression of P53 protein was higher in the model group than in the sham group (P<0.01), and was lower in the exercise preconditioning group than in the model group (P<0.01). Conclusion Exercise preconditioning coud down-regulate the expression of P53 protein in the ischemic penumbra, and inhibit the apoptosis of cortical cells.
