针刺对2型糖尿病大鼠弓状核瘦素受体基因的影响

Effect of Acupuncture on Leptin Receptor Gene Expression in Arcuate Nucleus of Type 2 Diabetes Mellitus Rats

摘要: 目的 探讨瘦素与弓状核瘦素受体(OB-R)基因表达和2型糖尿病(T2DM)的关系以及针刺对T2DM机体瘦素水平和弓状核OB-R基因表达的影响。方法 给食源性肥胖大鼠腹腔注射小剂量链脲佐菌素(STZ)造成T2DM模型,随机分为针刺组、优降糖组和模型组。处理4周后,用快速血糖仪检测空腹血糖(FBS),用放免法检测空腹胰岛素(FINS),用ELISA法检测空腹瘦素(FLP),用原位杂交检测弓状核OB-R mRNA表达,测量治疗前后大鼠的身长、体重,计算Lee's指数,并且与正常组比较。结果 模型组FBS、FINS高于正常组(P<0.05);治疗后,针刺组FBS、FINS明显下降(P<0.01),接近正常组水平;优降糖组FBS明显下降(P<0.01),FINS有所下降(P<0.05)。模型组FLP低于正常组(P<0.05)和优降糖组(P<0.01);优降糖组FLP高于正常组和针刺组(P<0.05)。模型组体重和Lee'S指数下降明显,针刺组、优降糖组体重和Lee's指数有所下降,接近正常组水平。模型组弓状核OB-R基因表达与正常组、针刺组、优降糖组无显著差异(均P>0.05),优降糖组弓状核OB-R基因表达明显低于正常组 (P<0.05),针刺组弓状核OB-R基因表达和正常组无显著性差异(P>0.05)。结论 T2DM大鼠模型明显消瘦后出现血清瘦素水平下降,优降糖组存在明显的瘦素抵抗,针刺可以在一定程度上改善弓状核OB-R基因表达。

Abstract: Objective To study the relationships among leptin, leptin receptor (OB-R) gene in arcuate nucleus and type 2 diabetes mellitus, and the effect of acupuncture on them. Methods Obese rats due to diet were injected in abdomen with small dosage streptozotocin to induce type 2 diabetes mellitus model, and were divided into acupuncture, glibenclamide and model group. Normal rats were in control. After treatment for 4 weeks, fasting blood sugar (FBS), fasting insulin (FINS), leptin, OB-R mRNA expression in arcuate nucleus were evaluated. Height, weight were measured and Lee's index were calculated before and after treatment. Results The FBS and FINS were higher in model group [(15.79±1.87) mmol/L, (37.20±3.92) mIU/L] than in control [(5.16±0.13) mmol/L, (23.57±1.63) mIU/L], but decreased in acupuncture group [(5.46±0.20) mmol/L, (20.87±1.80) mIU/L] and glibenclamide group [(5.26±0.13) mmol/L, (28.10±1.86) mIU/L]. Leptin was lower in model [(0.95±0.15) ng/ml] than in control [(1.32±0.15) ng/ml], but was higher in glibenclamide group [(1.83±0.23) ng/ml] than in control. Weight and Lee's index decreased in model, acupuncture and glibenclamide groups after treatment. OB-R mRNA expression in arcuate nucleus in model wasn't different from control, acupuncture or glibenclamide group, nor between acupuncture and control, but was lower in glibenclamide group than in control. Conclusion Leptin decreased in type 2 diabetes mellitus rats when they became thin. Leptin resistance appears when treated with glibenclamide. Acupuncture may improve the OB-R mRNA expression in arcuate nucleus to some extent.
