高频超声心动图对心肌梗死大鼠左心室重构的动态研究

Value of High Frequency Echocardiography to Evaluate Left Ventricular Remodeling in Rat with Acute Myocardial Infarction

摘要: 目的 探讨高频超声心动图对心肌梗死大鼠左心室重构评价的可行性。方法 结扎50只大鼠冠状动脉左前降支制备心肌梗死大鼠模型,术后24 h存活41只(82%),另选20只为假手术组。采用高频多普勒超声心动图观察术后2周、8周时心脏结构及功能,8周时进行血流动力学测定。结果 心肌梗死大鼠与假手术组比较,2周时梗死区室壁变薄,运动明显降低,左心室内径和容量增大,射血分数和缩短分数下降,二尖瓣舒张早期血流速度(E)增加,晚期血流峰值(A)降低,E/A比值增加;8周时梗死区斑痕化,心腔球形化,左心室内径和容量继续增大,左心室功能进一步下降,E/A比值继续增大。模型组左心室收缩末压力以及最大心室内压力变化速率下降,而左心室舒张末压力增加( P<0.05),左心室射血分数和缩短分数与dp/dtmax呈正相关。结论 高频超声心动图可以动态评价心肌梗死大鼠左心室的结构和功能。

Abstract: Objective To explore the value of high frequency echocardiography to evaluate cardiac structure and function of rat with acute myocardial infarction (AMI).Methods The rat AMI model was established by ligating the proximal left anterior descending coronary artery. The cardiac structure and function of model rats were examined with high frequency echocardiography in the 2nd and 8th week after ligation successfully. Another 20 healthy rat were selected as the sham operative group.Results The survival rat is 82.0% beyond 24 hours. At 2nd week, the model animals had an increased end-diastolic diameter (EDD), end-systolic diameter (ESD), end-diastolic volume (EDV), and further end-systolic volume (ESD), compared with the sham operative group. Ejection fraction (EF) and fraction shortening (FS) of model rats decreased. The ratio of E/A was higher. At 8th week, these parameters deteriorated left atrial diameter (LA) increased ( P<0.05). EF and FS were relevant to dp/dtmax.Conclusion High frequency echocardiography can evaluate left ventricle remodeling of rat dynamically.
