莫诺苷对大鼠局灶性脑缺血再灌注模型脑梗死体积的影响

Effect of Morroniside on Cerebral Infarction Volume in Focal Cerebral Ischemia-reperfusion Rats

摘要: 目的 观察莫诺苷对大脑中动脉阻塞法(MACO)致脑缺血再灌注模型大鼠脑梗死体积的影响。方法 用线栓法制备局灶性脑缺血再灌注大鼠模型,缺血30 min,再灌注7 d。采用2, 3, 5-氯化三苯基四氮唑(TTC) 染色,检测大鼠的脑梗死体积。结果 与假手术组相比,模型组及给药组出现明显梗死灶;与模型组相比,莫诺苷(90 mg/kg,270 mg/kg)脑梗死体积显著减少,维生素E(35 mg/kg)组脑梗死体积未出现明显改变。结论 莫诺苷可减少局灶性脑缺血再灌注大鼠脑梗死体积,对脑缺血再灌注损伤有保护作用。

Abstract: Objective To observe the effect of morroniside on cerebral infarction volume in focal cerebral ischemia-reperfusion rat model.Methods The animal model was induced by occlusion of middle cerebral artery with suture embolus, ischemia for 30 minutes, and reperfusion for 7 days. The infarction volume was measured by 2,3,5-triphenyltetrazolium chloride(TTC) staining technique.Results Compared with sham operation group, the cerebral infarction volume ratios increased obviously in model group and in the drug-treated(90 mg/kg,270 mg/kg)groups. Compared with model group, the cerebral infarction volume ratios decreased obviously in the morroniside-treated(90 mg/kg,270 mg/kg)group,while the cerebral infarction volume ratios in vitamin E-treated(35 mg/kg)group didn't change.Conclusion Morroniside may decrease the cerebral infarction volume after cerebral ischemia-reperfusion. It possesses protective effect against cerebral ischemia-reperfusion injury.
