中药筋脉通对高糖培养海马神经元细胞Bax、Bcl-2 和Caspase-3蛋白表达的影响

Effects of Jinmaitong on Expression of Bax, Bcl-2 and Caspase-3 in Hippocampal Neurons Cultured with High Glucose

摘要: 目的探讨中药筋脉通含药血清对高糖培养海马神经元细胞的保护作用。方法采用血清药理学方法制备筋脉通含药血清(JMT),原代培养新生鼠海马神经元细胞分别接种于不同条件的培养基中,分为正常对照组、高糖组、阳性对照组以及JMT大、中、小剂量组。不同培养基中培养72 h 后使用Western blotting 检测其Bax、Bcl-2 和Caspase-3 蛋白的表达。结果与正常对照组相比,高糖组Caspase-3、Bax 表达明显增加(P<0.01),Bcl-2 的表达下降(P<0.05);与高糖组相比,阳性对照组和JMT 各剂量组的Caspase-3、Bax 表达明显下降(P<0.01),Bcl-2 的表达升高(P<0.05);与阳性对照组相比,JMT 各剂量组的Caspase-3、Bax 表达下降(P<0.05),筋脉通大、中剂量组Bcl-2 的表达增加(P<0.05)。结论筋脉通含药血清可能通过提高Bcl-2 蛋白的表达,减少Bax、Caspase-3 的表达,上调Bcl-2/Bax 的比值,从而减少海马神经元细胞的凋亡。

Abstract: Objective To explore the effect of Jinmaitong (JMT) on expression of Bax, Bcl-2 and Caspase-3 in hippocampal neurons culturedwith high glucose. Methods Hippocampal neurons were primarily cultured and purified from the hippocampus of new-born SpragueDawley rats. Neuron-specific enolase immunocytochemical method was adopted for the identification of the neurons. They were divided intonormal control group, high glucose group, high-dose JMT (JH) group, middle-dose JMT (JM) group, low-dose JMT (JL) group and a positivecontrol group. 72 hours later, Western blotting was adopted to detect the expression of Bax, Bcl-2 and Caspase-3. Results Comparedwith the normal control group, the expression of Bax and Caspase-3 in the high glucose group significantly increased (P<0.01), and the expressionof Bcl-2 significantly decreased (P<0.05). Compared with the high glucose group, the expression of Caspase-3 and Bax in the positivecontrol group and JH, JM, and JL groups significantly decreased (P<0.01), and the expression of Bcl-2 significantly increased (P<0.05).Compared with the positive control group, the expression of Caspase-3 and Bax significantly decreased in JH, JM, and JL groups (P<0.05),and the expression of Bcl-2 significantly increased in JH and JM groups (P<0.05). Conclusion JMT may reduce apoptosis by inhibiting theexpression of Bax and Caspase-3 and promoting the expression of Bcl-2.
