红景天对大鼠脑缺血再灌注c-Fos表达及神经细胞凋亡的影响

Effects of Rhodiola on Expression of c-Fos and Neuronal Apoptosis after Cerebral Ischemia/Reperfusion Injury in Rats

摘要: 目的探讨红景天对大鼠脑缺血再灌注后脑组织中c-Fos 表达水平及神经细胞凋亡的影响。方法Wistar 大鼠120 只,分为假手术组、模型组和干预组(红景天),各组再分为3 h、6 h、12 h、24 h、48 h 共5 个亚组,每个亚组8 只大鼠。后两组采用大脑中动脉线栓法制备局灶性脑缺血再灌注模型,干预组术前红景天每天0.672 g/kg 灌胃15 d。参考Longa 法在大鼠麻醉清醒后进行评分,应用免疫组化、TUNEL法检测各组c-Fos 表达及神经细胞凋亡。结果模型组和干预组缺血脑组织中c-Fos 表达明显增加(P<0.01),于再灌注48 h 达高峰;与模型组相比,干预组c-Fos 表达减少(P<0.05),神经元凋亡明显减少(P<0.01),神经功能缺损评分下降(P<0.05)。结论红景天能抑制大鼠脑缺血再灌注后脑组织中c-Fos 表达,减少神经元凋亡,减轻缺血再灌注损伤。

Abstract: Objective To investigate the effect of Rhodiola Rosae P. E on the expression of c-Fos and the apoptosis of neuronal in rat brain after ischemia/reperfusion (I/R) injury. Methods 120 Wistar rats were divided into sham group, model group and intervention group,subgrouped as 3 h, 6 h, 12 h, 24 h, and 48 h. The model and intervention groups underwent middle cerebral artery occlusion, and the latter accepted Rhodiola 0.672 g/kg daily for 15 d. They were assessed with Longa's score. The expression of c-Fos and apoptosis of neuronal were measured with immunohistochemistry and TUNEL. Results The expression of c-Fos in the ischemic area increased after cerebral ischemia/reperfusion (P<0.01), peaking at 24 h after injury, and decreased in the intervention group (P<0.01), with the decrease of neuron apoptosis and Longa's score (P<0.01). Conclusion Rhodiola protects the brain tissue against ischemia/reperfusion injury by inhibiting the expression of c-Fos and apoptosis in rats.
