莫诺苷对局灶性脑缺血再灌注大鼠血管生成素1及其受体Tie-2 的影响

Effects of Morroniside on Expression of Angiopoietin-1 and Tie-2 in Rats after Focal Cerebral Ischemia-reperfusion

摘要: 目的观察莫诺苷对局灶性脑缺血再灌注大鼠血管生成素1(Ang-1)及其受体Tie-2 的影响。方法20 只健康成年雄性Sprague-Dawley大鼠随机分为假手术组,模型组,莫诺苷小、中、大剂量组,每组4 只。线栓法制备大鼠大脑中动脉阻塞模型,术后予莫诺苷30 mg/kg、90 mg/kg、270 mg/kg 每天1 次灌胃。术后7 d 采用Western blotting 法分析皮层Ang-1 及其受体Tie-2 的表达。结果模型组患侧皮层Ang-1、Tie-2 的表达均比假手术组明显增加(P<0.01)。莫诺苷大剂量组Ang-1、Tie-2 的表达较模型组明显提高(P<0.01),莫诺苷中、大剂量组Tie-2 的表达较模型组显著提高(P<0.001)。结论莫诺苷能上调局灶性脑缺血大鼠缺血再灌注皮层Ang-1及受体Tie-2 的表达,促进血管再生。

Abstract: Objective To explore the effects of morroniside on the expression of Angiopoietin-1 (Ang-1) and Tie-2 in a rat after focal cerebral ischemia-reperfusion. Methods 20 male Sprague-Dawley rats were randomly divided into sham group (n=4), ischemia group (n=4), and morroniside groups (low, medium and high dosage groups, n=4). The middle cerebral artery were occluded for 30 min, and re-perfused. Morroniside was administered intragastrically once a day at dose of 30 mg/kg, 90 mg/kg and 270 mg/kg after operation. The expression of Ang-1 and Tie-2 in the ischemic ipsilateral cortex were detected with Western blotting analysis 7 days after operation. Results The expression of Ang-1 and Tie-2 increased in the ischemia group compared with the sham group (P<0.01), and both of them further increased in the morroniside groups of high dosage compared with the ischemia group (P<0.01), and the expression of Tie-2 also increased in the morroniside groups of medium dosage (P<0.001). Conclusion Morroniside could increase the expression of Ang-1 and Tie-2 in the ischemic ipsilateral cortex after ischemia-reperfusion in rats, suggesting promoting the angiogenesis after ischemia.
