小鼠脊髓损伤后基质金属蛋白酶-9及血脊髓屏障的变化

Alteration of Blood-spinal Cord Barrier and Matrix Metalloproteinases-9 in Mice after Spinal Cord Injury

摘要: 目的 探讨脊髓损伤后基质金属蛋白酶-9(MMP-9)表达变化对血脊髓屏障(BSCB)通透性的影响。方法 C57BL/6小鼠(n=68)随机分为假手术组、损伤后2 d组(S2)、损伤后7 d组(S7)和损伤后14 d组(S14),每组17只。损伤组采用改良Allen's法制作中度脊髓损伤模型。在损伤后的第2、7、14天分别对小鼠进行盲法的小鼠后肢运动功能(BMS)评分和BMS亚评分;各组小鼠腹腔注入伊文思蓝,检测 BSCB通透性;采用免疫荧光法检测紧密连接蛋白(occludin)表达,采用免疫印迹法检测 occludin和 MMP-9表达。结果 术后 2 d BMS评分显著降低,BSCB通透性显著增高;术后 14 d BMS评分升高(P<0.01),BSCB通透性下降(P<0.05)。术后7 d,occludin表达下调,MMP-9表达上调(P<0.05);术后14 d occludin表达上调,MMP-9表达下调(P<0.05)。结论 脊髓损伤后MMP-9可能通过介导occludin表达,影响BSCB通透性改变。

Abstract: Objective To investigate the influence of matrix metalloproteinases-9 (MMP-9) on permeability of blood-spinal cord barrier (BSCB) after spinal cord injury (SCI). Methods 68 male C57BL/6 mice were randomly divided into sham group, 2 days group (S2), 7 days group (S7) and 14 days group (S14) after SCI with 17 mice in each group. All the groups received a moderate impacted spinal cord injury except the sham group. Evan's Blue (EB) was administered intraperitoneally to detect the permeability of BSCB. Occludin was analyzed by immunofluorescence, the expressions of occludin and MMP-9 were detected by Western blotting. Results After SCI, BMS score significantly reduced, compared with S2 group, S14 group showed a significant increase (P<0.01). The permeability of BSCB was seriously damaged after SCI. Compared with S2 group, S14 group showed a notable down-regulation in the permeability of injured micro-vessels (P<0.05). The expression of occludin was down-regulated and the expression of MMP-9 was up-regulated 7 days after SCI (P<0.05). Compared with S2 group, S14 group showed a significant up-regulation of occludin and a remarkable down-regulation of MMP-9 (P<0.05). Conclusion After SCI, MMP-9 might mediate the expression of occludin to influence the BSCB permeability.
