黄芪多糖对糖尿病鼠胰岛IL-4、IFN-γ的影响

Effect of Astragalus Polysaccharides on Expression of IL-4 and IFN-γ in Non-obese Diabetic Mice

摘要: 目的探讨黄芪多糖(APS)对非肥胖糖尿病(NOD)小鼠胰岛白细胞介素-4(IL-4)、干扰素γ(IFN-γ)表达的影响。方法20只NOD小鼠随机分为APS组和对照组,各10只。APS组小鼠予APS2g/kg/d口服2个月,对照组小鼠予等量生理盐水。比较两组动物的糖尿病发病率、胰岛组织病理和IL-4、IFN-γ表达的变化。结果与对照组相比,APS组NOD鼠1型糖尿病发病率明显下降,发病时间延缓,胰岛组织保持完好,IFN-γ表达下调,IL-4表达上调。结论APS可能有纠正1型糖尿病的细胞因子免疫失衡的作用。

Abstract: Objective To investigate the effect of astragalus polysaccharide(APS)on expression of interleukin-4(IL-4)and interferon-γ(IFN-γ)in non-obese diabetic(NOD)mice.Methods 20 NOD mice were randomly divided into the APS group and control group with 10 animals in each group.The mice of the APS group took APS 2 g/kg/d orally for two months.The animals of the control group took same quantity saline.The incidence rate of type 1 diabetes mellitus of two groups was calculated,and pancreatic histopathology was observed with microscope,and expression of IL-4 and IFN-γ were examined with RT-PCR for both APS group and control group.Results The incidence rate of type 1 diabetes mellitus of the APS group was lower than the control group,and histopathology of islets were more protected,expression of IFN-γ was lower and expression of IL-4 was higher in the APS group.Conclusion APS may correct the imbalance of cytokines.
